Description

Humanin is a mitochondrial-encoded 24-amino acid peptide known for its cytoprotective and neuroprotective properties. It represents one of the first identified mitochondrial-derived peptides (MDPs), functioning as an intracellular and extracellular signaling molecule involved in cell survival, apoptosis inhibition, and metabolic regulation.

Humanin exerts its effects through multiple receptor pathways, including formyl peptide receptor-like 1 (FPRL1) and CNTF receptor/gp130/WSX-1 complexes, leading to activation of STAT3, ERK1/2, and PI3K/Akt signaling.
Preclinical studies indicate roles in protecting neurons from oxidative stress, modulating insulin sensitivity, and suppressing pro-apoptotic factors such as Bax.

Because of its broad systemic protective effects, Humanin serves as a valuable model for research into mitochondrial signaling, aging biology, metabolic disorders, and neurodegenerative disease mechanisms.
