Description

PNC-27 is a synthetic chimeric peptide engineered to selectively induce cell-membrane disruption in cancer cells by targeting HDM-2 (Human Double Minute-2) — a negative regulator of the p53 tumor-suppressor protein.

The peptide contains an HDM-2–binding domain derived from p53 (residues 12–26) fused to a cell-penetrating leader sequence (membrane-active segment of MRP).
Preclinical research has shown that PNC-27 binds HDM-2 expressed on the plasma membranes of malignant cells, forming transmembrane pores that lead to selective necrosis or apoptosis of tumor cells, while sparing normal, non-HDM-2–expressing cells.

Because of this selectivity, PNC-27 is widely used in oncology and membrane biophysics research as a model for targeted peptide-induced cytotoxicity, p53 pathway restoration, and HDM-2–mediated cancer biology.
